Multiphoton imaging of the effect of monosaccharide diffusion and formation of fluorescent advanced end products in porcine aorta

Prolonged exposure of tissues to elevated blood sugar levels lead the formation advanced glycation end products (AGEs), thus contributing diabetic complications. Since vascular system is in immediate contact with blood, effects on aorta a major health concern. However, relative effect diffusion molecular through wall and rate AGE not known. In this study, we aim address issue by incubating excised porcine D-glucose, D-galactose, D-fructose solutions for different periods. The tissue specimens were then multiphoton imaging autofluorescence intensity profiles across wall. We found that Days 4 48 incubation, constant along radial direction sections, suggesting monosaccharide rapid comparison fluorescent AGEs (fAGEs). Moreover, aorta, fAGE D-glucose are factors 2.08 1.14 D-galactose. Our results suggest prolonged cardiovascular monosaccharides days or longer, damage uniform throughout tissues.
